ABSTRACT

Introduction: Malathion is a pesticide that is metabolized in the body into malaoxon and is free radical. Malaoxon works to inhibit the acetylcholinesterase enzyme and free radicals causing oxidative stress that can affect liver levels of SOD and MDA. U. lobata leaf decocta has antioxidant, anti-inflammatory and hepatoprotective effects, however research on the effects of U. lobata leaf decocta on levels of SOD and MDA is still rare. This study aims to prove the effect of U. lobata leaf decocta on levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in the liver of Danio rerio exposed to chronic malathion.

Method: In vivo laboratory experimental design with control group post test only. Zebrafish was divided into 2 control groups and 3 treatment groups giving decoction of U. lobata leaves at a dose of 125 mg / L, 250 mg / L, 500 mg / L.

SOD and MDA levels were measured using spectophotometry with wavelengths of 450 nm and 532 nm, respectively. Data analysis using One way ANOVA followed by LSD test with a significant level of p <0.05.

Result: Decoction of urena lobata leaves at a dose of 125 mg / L, 250 mg / L, 500 mg / L inhibited the decrease in superoxide dismutasae (SOD) levels, respectively 30%, 90% and 70% compared to the control group (p <0.05), and can inhibit the increase in malondialdehyde (MDA) levels respectively 40%, 60% and 55% compared to the control group.

Conclusion: Urena lobata leaf decoction inhibited the decrease in SOD levels and inhibited the increase in liver MDA levels in Danio rerio exposed to chronic malathion for 40 days.

Pendahuluan: Malathion merupakan pestisida yang dimetabolisme didalam tubuh menjadi malaoxon  dan bersifat radikal bebas. Malaoxon bekerja menghambat enzim asetilkolinesterase dan radikal bebas menyebabkan stress oxidative sehingga dapat mempengaruhi kadar SOD dan MDA hepar. Dekota daun U. lobata mempunyai efek sebagai antioksidan, antinflamasi dan hepatoprotektor, namun penelitian tentang efek dekokta daun U. lobata terhadap kadar SOD dan MDA hepar masih jarang dilakukan. Penelitian ini bertujuan untuk membuktikan pengaruh dekota daun U. lobata terhadap kadar superoxide dismutase (SOD) dan malondialdehyde (MDA) hepar Danio rerio yang dipapar malathion kronik

Metode: Eksperimental laboratorium in vivo dengan desain control group post tes only. Ikan zebra dibagi menjadi  2 kelompok kontrol dan 3 kelompok perlakuan pemberian dekokta daun U. lobata dosis 125 mg/L, 250 mg/L, 500 mg/L.

kadar SOD dan MDA diukur menggunakan spektofotometri dengan panjang gelombang berturut turut 450 nm dan 532 nm. Analisa data menggunakan One way ANOVA dilanjutkan dengan uji LSD dengan tingkat signifikan p<0,05.

Hasil: Dekokta daun urena lobata pada dosis 125 mg/L, 250 mg/L, 500 mg/L menghambat penurunan kadar superoxide dismutasae (SOD) berturut-turut 30%, 90% dan 70% dibandingkan kelompok kontrol (p<0,05), serta dapat menghambat peningkatan kadar malondialdehyde (MDA) secara berturut-turut 40%, 60% dan 55% dibandingkan kelompok kontrol.

Kesimpulan: Pemberian dekokta daun Urena lobata menghambat penurunan kadar SOD dan menghambat peningkatan kadar MDA hepar pada Danio rerio yang dipapar malathion kronik 40 hari.
